Sarcomeric versus Non-Sarcomeric HCM

چکیده
Hypertrophic cardiomyopathy (HCM) is the most common heritable cardiovascular disorder and characterized by left ventricular hypertrophy (LVH), which unexplained abnormal loading conditions. HCM inherited as an autosomal dominant trait and, in about 40% of patients, causal mutation identified genes encoding sarcomere proteins. According to results genetic screening, patients are currently categorized two main sub-populations: sarcomeric-positive (Sarc+) whom a sarcomeric gene; sarcomeric-negative (Sarc−) has not been identified. In rare cases, Sarc− cases may be caused pathogenic variants non-sarcomeric genes. The aim this review describe differences phenotypic expression clinical outcomes Sarc+ briefly discuss current knowledge mutations.

Erratum to: Early Results of Sarcomeric Gene Screening from the Egyptian National BA-HCM Program
The present study comprised sarcomeric genotyping of the three most commonly involved sarcomeric genes: MYBPC3, MYH7, and TNNT2 in 192 unrelated Egyptian hypertrophic cardiomyopathy (HCM) index patients. Mutations were detected in 40 % of cases. Presence of positive family history was significantly (p=0.002) associated with a higher genetic positive yield (49/78, 62.8 %). The majority of the de...
متن کاملSarcomeric proteins and inherited cardiomyopathies.
Over the last two decades, a large number of mutations have been identified in sarcomeric proteins as a cause of hypertrophic, dilated or restrictive cardiomyopathy. Functional analyses of mutant proteins in vitro have revealed several important functional changes in sarcomeric proteins that might be primarily involved in the pathogenesis of each cardiomyopathy. Creation of transgenic or knock-...
متن کاملOxidative stress and sarcomeric proteins.
Oxidative stress accompanies a wide spectrum of clinically important cardiac disorders, including ischemia/reperfusion, diabetes mellitus, and hypertensive heart disease. Although reactive oxygen species (ROS) can activate signaling pathways that contribute to ischemic preconditioning and cardioprotection, high levels of ROS induce structural modifications of the sarcomere that impact on pump f...
متن کاملPrediction and visualization data for the interpretation of sarcomeric and non-sarcomeric DNA variants found in patients with hypertrophic cardiomyopathy.
Genomic technologies are redefining the understanding of genotype-phenotype relationships and over the past decade, many bioinformatics algorithms have been developed to predict functional consequences of single nucleotide variants. This article presents the data from a comprehensive computational workflow adopted to assess the biomedical impact of the DNA variants resulting from the experiment...
متن کاملSarcomeric dysfunction in heart failure.
Sarcomeric dysfunction plays a central role in reduced cardiac pump function in heart failure. This review focuses on the alterations in sarcomeric proteins in diseased myocardium that range from altered isoform expression to post-translational protein changes such as proteolysis and phosphorylation. Recent studies in animal models of heart failure and human failing myocardium converge and indi...
